fa بررسی اثرهای تعدیل‌کنندگی ایمنی وزیکول‌های خارج‌سلولی و مایع فاقد سلول باکتری پروبیوتیک باسیلوس کوآگولانس همر بر روی ردۀ سلولی سرطان رودۀ بزرگ HT-29 Investigating the immunomodulatory effects of extracellular vesicles and cell-free liquid of the probiotic bacteria Bacillus coagulans Hammer on colon cancer cell line HT-29 پروبیوتیک Probiotic مقاله پژوهشی Research Article چکیده سابقه و هدف: باکتری باسیلوس کوآگولانس همر (Bacillus coagulans Hammer) یک گونۀ پروبیوتیک غیرپاتوژن و گرم‌مثبت است که قادر به آزادسازی وزیکول‌های خارج‌سلولی می‌باشد. این وزیکول‌ها حاوی پروتئین‌های سطحی خودِ باکتری می‌باشند که قادر به تحریک سیستم‌ایمنی هستند. در نوزادان و افراد دارای نقص سیستم‌ایمنی استفاده از این وزیکول‌های خارج‌سلولی در مقایسه با باکتری زنده، خطر بسیار کمتری دارد. در نتیجه استفاده از این وزیکول‌ها باعث ایجاد تغییر و بهبود در پاسخ‌های التهابی می‌شوند. در این مطالعه هدف ما بررسی تغییرهای ایجادشده در پاسخ‌های ایمنیِ بعد از استفاده از وزیکول‌های خارج‌سلولی و مایع فاقدسلولِ باکتریِ B. coagulans Hammer بر روی سلول‌های HT-29 به‌‌عنوان ردۀ سلولیِ سرطان رودۀ بزرگ می‌باشد. مواد و روش‌ها: در این مطالعه وزیکول‌های مشتق‌شده از باکتری B. coagulans Hammer با استفاده از پلی‌اتیلن‌گلایکول استخراج شدند. مایع فاقدسلول باکتری آماده‌سازی شد و سلول‌های HT-29 با غلظت 10 میکروگرم در میلی‌لیتر وزیکول‌های مشتق‌شده و رقّت 20 درصد حجمی‌حجمی‌ مایعِ رویی فاقدسلول تیمار گردید. روش RT-qPCR جهت بررسی میزان بیان‌ژن‌های التهابی IL-1β،IL-6 ،IL-8،IL-10 ،TNF-α  و  TGF-βو نیز روش اِلایزا برای بررسی میزان سایتوکاین‌های TGF-β و TNF-α تولیدشده مورداستفاده قرار گرفت. یافته ها: نتایج به‌دست‌آمده از RT-qPCR نشان داد که وزیکول‌های مشتق‌شده و مایعِ فاقدسلول باکتری به‌خوبی قادر به کاهش بیان‌ژن‌های التهابی IL-1β ، IL-8،IL-6   و TNF-α و افزایش بیان‌ژن‌های غیرالتهابی IL-10،  TGF-βشده‌اند. بررسی سایتوکاین‌ها به‌روش اِلایزا نیز نشان‌دهندۀ افزایش TGF-β و کاهش TNF-α در سلول‌های تیمارشده است. نتیجۀ گیری: نتایج به‌دست‌آمده از مطالعه حاضر نشان داد وزیکول‌ها و مایع فاقدسلولِ باکتری B. coagulans Hammer علاوه‌بر کاهش پاسخ‌های التهابی قادر به‌ افزایش پاسخ‌های غیرالتهابی نیز هستند.   Abstract Aim and Background: Bacillus coagulans Hammer is a Gram-positive probiotic that releases extracellular vesicles (EV). These vesicles contain the surface proteins of the bacteria, which are able to stimulate the immune system. For newborns or immunosuppressed people, the use of these extracellular vesicles is much less dangerous. As a result, the use of these vesicles causes a change and improvement in the inflammatory responses. In this study, our aim is to investigate the changes in immune responses after using extracellular vesicles and cell-free liquid of B. coagulans Hammer on HT-29 cells, as a colon cancer cell line. Materials and Methods: B. coagulans hammer EVs were extracted using polyethylene glycol. Cell-free supernatant was prepared, and HT-29 cells were treated with a concentration of 10 μg/mL of EVs and a dilution of 20% v/v. The RT-qPCR method was used to assess the expression levels of inflammatory genes IL-1β, IL-6, IL-8, IL-10, TNF-α, and TGF-β, and the ELISA method was used to assess the levels of TGF-β and TNF-α cytokines produced in the case. Results: The results obtained from RT-qPCR showed that the EVs and Cell Free Supernatant can reduce the expression of inflammatory genes IL-1β, IL-6, IL-8, TNF-α and increase the expression of anti-inflammatory genes IL-10 and TGF-β. TNF-α and TGF-β cytokines have been examined by the ELISA method also shows an increase in TGF-β and a decrease in TNF-α in the treated cells.
