سابقه‌ و هدف: در دو دهه گذشته، پروتئین‌ها و پپتیدها تبدیل به یک گروه مهم از داروها شده‌اند. با این حال، حساسیت به تخریب شیمیایی و فیزیکی از چالش‌های اصلی کاربردهای صنعتی و دارویی پروتئین‌ها است. کمابیش90 درصد از وزن پرمرغ از ضایعات صنایع دام و طیور، کراتین‌ است. این پروتئین نامحلول بوده و تمایل شدیدی به تجمع و رسوب‌دهی دارد.
مواد و روش‌ها: در این پژوهش کراتین از پر مرغ با محلول سدیم هیدروکسید 2/0 مولار استخراج شد و توسط آنزیم آلکالاز صنعتی (V/V) 8 درصد در دمای 55 درجه‌ سانتی‌گراد به­مدت 5 ساعت در pH برابر 8 هیدرولیز شد. محلول کراتین به­دست آمده، توسط روش آلکیلاسیون و سولفیدولیز به­منظور کاهش اگریگاسیون مورد اصلاح ساختار شیمیایی قرار گرفت. حلالیت کراتین، درجه اصلاح شیمیایی، درصد پایداری، مقدار مول تیول آزاد و هم­چنین DLS و FTIR نمونه‌ها بررسی شد.
یافته‌ها: نتایج نشان داد درجه‌ اصلاح شیمیایی کراتین در روش آلکیلاسیون 72 درصد و در روش سولفیدولیز با 66 درصد بوده است. اصلاح شیمیایی کراتین هیدرولیز شده باعث کاهش اگریکاسیون در 3pH به­میزان 65/288 درصد و 3/335 درصدی به­ترتیب برای روش آلکیلاسیون و روش سولفیدولیز به­دست آمده‌است. بررسی پایداری و حفظ محتوی پروتئینی نشان داد، کراتین اصلاح شده به­روش آلکیلاسیون 9/81 درصد و کراتین اصلاح شده به روش سولفیدولیز 3/86 درصد در طی 30 روز پایدار بوده‌است در صورتی­که کراتین هیدولیز شده به­عنوان شاهد تنها 8/9 درصد از محتوای پروتئینی خود را حفظ کرده و پایداری داشته‌است. نتیجه‌ تست DLS نمونه‌ محلول کراتین اصلاح شده با روش آلکیلاسیون در آب، دارای  476/0= PI و اندازه متوسط ذرات 3/157 نانومتر است، در نمونه‌ کراتین اصلاح شده با روش سولفیدولیز در آب، مقدار 475/0=PI و اندازه‌ متوسط ذرات 2/136 نانومتر تعیین گردید، که نسبت­به نمونه‌ کراتین هیدرولیز شده با 552/0=PI و اندازه‌ متوسط 3/990 نانومتر، یکنواخت‌تر و پراکنده‌تر بوده است.
نتیجه‌گیری: اصلاح شیمیایی انجام شده بر روی کراتین هیدولیز شده تغییری دائمی بوده و اتصال گروه‌های عاملی مورد انتظار باعث افزایش حلالیت کراتین در محیط‌های آبی و کاهش تجمع در pH‌ اسیدی شده است. هم­چنین نتایج آنالیز FTIR اصلاح شیمیایی کراتین را تأیید کرد.
