fa ارزیابی مولکولی هرپس سیمپلکس ویروس 2 در سقط های ایدیوپاتیک به روش PCR PCR Detection of Herpes Simplex Virus 2 in Idiopathic Abortions سلولی و مولکولی Cellular and molecular مقاله پژوهشی Research Article چکیده سابقه و هدف: ویروس هرپس سیمپلکس نوع 2(HSV-2 ) از مهمترین ویروسهای بیماریزای انسانی است. این ویروس باعث تبخال تناسلی شده و در زنان ممکن است باعث سقط جنین، عوارض بارداری و کاهش باروری شود. هدف این مطالعه تشخیص سریع HSV-2 در سقطهای ایدیوپاتیک بهروش مولکولی PCR است. مواد و روش ها: 50 نمونه سرم خون زنان با سقطهای ایدیوپاتیک جمع آوری و به آزمایشگاه منتقل گردید. آزمون PCR با استفاده از DNA سوش استاندارد HSV-2 بهینه و سپس حد تشخیص (LOD) و ویژگی تست با استفاده از DNA عوامل مختلف بررسی گردید. آزمون استاندارد با استفاده از کنترل مثبت و منفی بر روی نمونه ها انجام و نتایج بهروش آگاروز ژل الکتروفورز بررسی گردید. یافته ها: تست PCR بهینه و محصول bp 231 در ژل آگاروز مشاهده گردید. در آزمون ویژگی فقط با DNA ی  HSV-2نتایج مثبت حاصل و همین طور حد تشخیص  Copy/Reaction10به دست آمد. از 50 نمونه مورد بررسی، 9 نمونه (5/4 %) مثبت گردید. نتیجه گیری: سقط های ایدیوپاتیک، سقط هایی هستند که به ظاهر عامل مشخص ندارند. اما به طور مشخص عوامل متعددی از جمله عوامل عفونی که شناسایی آنها مشکل است، می توانند موجب سقط جنین گردند. یکی از این عوامل می تواندHSV-2  باشد. نتایج این مطالعه مشخص کرد که در این نوع سقط ها HSV-2 یکی از عوامل مهم است و با آزمون هایی مانند PCR به سرعت میتوان، HSV-2 را شناسایی نمود. Abstract Aims and Background: Herpes simplex virus type 2 (HSV-2) is one of the most important human pathogenic viruses. The virus causes genital herpes and can cause miscarriage, pregnancy complications, and infertility in women. The aim of this study was to evaluate the role of HSV-2 in idiopathic abortion by PCR. Materials and Methods: Fifty blood serum samples of women with idiopathic miscarriages were collected and transferred to the laboratory. The PCR test was performed using standard HSV-2 strain DNA and then the limit of detection (LOD) and specificity of the test was determined using a different strain of DNA. A standard test was performed using positive and negative controls on the samples and the results were analyzed by agarose gel electrophoresis. Results: The PCR technique est optimized and 231 bp amplicon were observed in agarose gel. In specificity test only with DNA HSV-2 achieved positive results as well as a limit of detection 10 Copy / Reaction. Of the 50 samples studied, 9 (4.5%) were positive. Conclusion: Idiopathic miscarriages are miscarriages that do not seem to have a definite cause. But clearly, several factors, including infectious agents that are difficult to identify, can cause miscarriage. One of these factors could be HSV-2.
