fa تکنیک واکنش زنجیره ای پلیمراز (PCR) برای تشخیص سریع سیتومگالوویروس در سقط های ایدیوپاتیک Polymerase Chain Reaction (PCR) technique for rapid detection of Cytomegalovirus in idiopathic abortions میکروبیولوژی Microbiology مقاله پژوهشی Research Article سابقه و هدف: سایتومگالوویروس (CMV) انسانی از اعضای خانواده ی هرپس ویروس ها است که از علل اصلی عفونت ویروسی داخل رحمی  می باشد. هدف این مطالعه  بررسی تشخیص CMV در زنان با سقط های ایدیوپاتیک توسط تکنیک واکنش زنجیره پلیمراز (PCR)  می باشد. مواد و روش ها: 50 نمونه خون از زنان مبتلا به سقط های ایدیوپاتیک جمع آوری و توسط کیت تجاری   DNG-Plus ژنوم  ویروسی استخراج گردید. تکنیک PCR با استفاده از ژنوم سویه استاندارد CMV بهینه سازی شد و سپس تعیین حد تشخیص (LOD) و اختصاصیت آزمایش با استفاده از ژنوم های غیر از CMV مورد ارزیابی قرار گرفت. تکنیک PCR بهینه شده بر روی نمونه ها و کنترل های مثبت و منفی انجام شد و نتایج توسط روش الکتروفورز ژل مورد تجزیه و تحلیل قرار گرفت. یافته ها: با استفاده از تکنیک PCR بهینه شده، محصول 257 جفت بازی در ژل آگاروز مشاهده گردید. در آزمون تعیین اختصاصیت، نتایج مثبت فقط با ژنوم CMV  حاصل شد و همینطور حد تشخیص 100کپی در هر واکنش بدست آمد. در نهایت، از 50 نمونه ی مورد بررسی، دو نمونه از نظر وجود CMV مثبت گردید. نتیجه گیری: سقط های ایدیوپاتیک، سقط هایی هستند که ظاهرا عامل مشخص ندارند. اما مشخصا عوامل متعددی از جمله عوامل عفونی که شناسایی آنها مشکل می باشد می توانند موجب سقط جنین گردند. یکی از این عوامل می تواند CMV باشد و نتایج این مطالعه مشخص کرد که بهتراست در این نوع سقط ها توسط آزمون های مولکولی مانند PCR به سرعت، CMV را جستجو و شناسایی نمود. Aim and Background Cytomegalovirus (CMV) is a member of the herpesviridae family, which is a major cause of intrauterine viral infection. The aim of this study was to evaluate the diagnosis of CMV in women with idiopathic abortions by the polymerase chain reaction (PCR) technique. Materials and Methods 50 blood samples of women with idiopathic abortions were collected and viral genome was extracted using commercial kit DNG-Plus. The PCR technique was optimized using the standard CMV strain genome and then the Limit of Detection (LOD) and specificity of the test was evaluated using genomes other than CMV. The optimized PCR technique was performed on the samples and positive and negative controls and the results were analyzed by gel electrophoresis. Results using optimized PCR technique, 257bp product were observed in agarose gel. In the specificity test, positive results were obtained only with the CMV genome and a detection limit of 100 copies per reaction was obtained. Finally, out of 50 samples, two samples were positive for CMV. Conclusion Idiopathic abortions are abortions that do not seem to have a specific cause. But obviously many factors including infectious agents that can cause abortion are difficult to identify them.
