fa مقایسه بیان miR-509-3-5pوmiR-596در بافت توموری و بافت حاشیه تومور در بیماران مبتلا به سرطان پستان Investigating the expression of miR-509-3-5p and miR-596 in tumor tissue and tumor peripheral tissue in breast cancer patients فیزیولوژی Physiology مقاله پژوهشی Research Article چکیده سابقه و هدف:سرطان پستان رایج ترین نوع سرطان در زنانمیباشد که در بافت اپیتلیال غده پستان ایجاد می‌شود. عوامل مختلف محیطی، ژنتیکی و اپی ژنتیکی دربروز سرطان پستان نقشدارند. طبق مطالعات انجام شده یکی از مهمترین تغییرات اپیژنتیکی درگیر در بروز سرطان پستان، تغییر در میزان بیان microRNAها میباشد. بنابراین هدف از این مطالعه بررسی بیوانفوماتیکی و آزمایشگاهی میزان بیان miR-509-3-5pو miR-596در بافت توموری سرطان پستان میباشد. مواد و روش ها:در تجزیه و تحلیل بیوانفورماتیکی مجموعه دادههای مربوط به miRNAها (GSE40525 و GSE45666) از پایگاه داده GEOتهیه شدهاند. تجزیه و تحلیل دادهها با استفاده ازپکیج  Affy در نرم‌افزار Rانجام شد.در مطالعات آزمایشگاهی از 100نمونه بافت تومور مبتلا به سرطان پستان و 100 نمونه بافت حاشیه تومور  با اخذ رضایت نامه کتبی نمونه برداری انجام شد. استخراج RNA بافتی با استفاده از محلول ترایزول انجام شد. سپس با استفاده از کیت سنتز cDNA از روی RNAهای استخراج شده DNA سنتز شد، و با استفاده از روش Real-time PCR میزان بیان miR-509-3-5p و miR-596 مورد بررسی قرار گرفت. یافته ها:با توجه به نتایج بیوانفورماتیکی میزان بیان miR-509-3-5p و miR-596 در بافت سرطان پستان نسبت به بافت سالم کاهش پیدا میکند. نتایج مربوط به بررسی بیان miR-509-3-5p و miR-596 در نمونههای توموری و سالم نیز نشان داد که سطح بیان miR-509-3-5p و miR-596 در بافت توموری نسبت به بافت سالم کاهش معنی داری را نشان میدهد (0.05(p<که این نتایج تایید کننده نتایج بررسیهای بیوانفورماتیکی بود. نتیجه گیری:با توجه به نتایج حاصل از مطالعه حاضر که نشان دهندهی کاهش بیان miR-509-3-5p و miR-596 در سرطان پستان بود. میتوان گفت که از این miRNAها میتوان به عنوان نشانگرهای زیستی تشخیصی و درمانی مهمی در تشخیص، پیش آگهی و درمان سرطان پستان استفاده کرد.   Abstract Aim and Background: Breast cancer is the most common type of cancer among women, which occurs in the epithelial tissue of the mammary gland. Various environmental, genetic and epigenetic factors play a role in the occurrence of breast cancer. According to the previous studies, one of the most important epigenetic changes involved in the occurrence of breast cancer is the dysregulationof the expression level of microRNAs. Therefore, the aim of this study was to investigate the expression level of miR-509-3-5p and miR-596 in breast cancer tumor tissue. Material and methods:In the bioinformatics analysis, the datasets related to miRNAs (GSE40525 and GSE45666) were prepared from the GEO database. Data analysis was performed using the Affy package in R software. Sampling was done from 100 women with breast cancer and 100 control. Tissue RNA was extracted using Trizol solution. Then, using the cDNA synthesis kit, DNA was synthesized from the extracted RNAs, and the expression level of miR-509-3-5p and miR-596 was investigated using Realtime PCR method. Results:According to the bioinformatics results, the expression level of miR-509-3-5p and miR-596 in breast cancer tissue reduced compared to healthy tissue. The expression level of miR-509-3-5p and miR-596 in tumor tissue was significantly lower than normal tissue (p<0.05).So these results confirmed the results of bioinformatics studies.
