fa طراحی و ساخت آدنوویروس های نوترکیب انکولیتیک به منظوردرمان رده های سلولی سرطان معده: رویکرد مبتنی بر وایرال وکتور Design and construction of recombinant oncolytic adenoviruses for the treatment of gastric cancer cell lines: a viral vector-based approach فیزیولوژی Physiology مقاله پژوهشی Research Article سابقه و هدف: ویروس درمانی انکولیتیک به‌عنوان راهکاری نویدبخش برای درمان سرطان شناخته میشود. ویروسهای انکولیتیک مانند آدنوویروس‌ها قادرند در سلولهای سرطانی تکثیر و آن‌ها را متلاشی کنند. آدنوویروس سروتیپ 5 ازطریق گیرنده کوکساکی- آدنوویروس به سطح سلولهای میزبان متصل میشود. میزان بیان این گیرنده در سطح سلولهای سرطانی بسیار متغییر است. هدف از این مطالعه، ساخت آدنوویروس انکولیتیک با ویژگی انتخابی بالا برای سلولهای آدنوکارسینومای معده(AGS) بوده که قادرند در این سلولها تکثیر و آنها را تخریب کنند.   مواد و روش ها: در مطالعه تجربی حاضر، توالی اکتروتات در ناحیهHI-loop  فیبرآدنوویروس کلون گردید و بدین ترتیب وکتورهای نوترکیب  pAd5TATE(wt)و pAd5TATE(ΔE1,ΔE3) ساخته شدند. وکتور pAdZ-TATE(wt) با استفاده از سیستم کلونینگ pADZ سنتز شد. ذرات ویروسی نوترکیب Ad5TATE(wt) و Ad5TATE(ΔE1,ΔE3) سنتز شد و در سلول های HEK293 تکثیر شد. با استفاده از روش کلسیم فسفات، وکتورهای نوترکیب به رده سلولیHEK293  منتقل و بدین ترتیب ذرات ویروسی جدید تولید شد. رده سلولی AGS با ویروس Ad5(wt) و Ad5TATE(wt) آلوده شدند و در زمانهای مختلف، بروز اثرات تخریب سلولی در این سلولها بررسی شد. با استفاده از آزمونMTT، میزان بقای سلولی در رده سلولی AGS تیمار شده با ویروس نوترکیب بررسی گردید. یافته ها: اثرات تخریب سلولی(CPE) در کمتر از 18 ساعت شروع و 72 ساعت بعد از آلودگی کامل گردید. تیتر ویروسهای نوترکیب با روش TCID50 برابر107 (برای Lµ  100 از محلول ویروسی) محاسبه شد. همچنین تیتر ویروس Ad5TATE(ΔE1,ΔE3) با روش TCID50 برابر TCID50/ml 107 بود. نتایج نشانداد، Ad5TATE(wt) در مقایسه باAd5(wt) ، توانایی آلوده‌سازی سلولهای رده AGS را دارد. نتایج حاصل از آزمون MTT نشان ‌داد، با افزایش غلظت ذرات ویروسی و زمان نگهداری، تعداد سلولهای زنده AGS به‌طور قابل‌ملاحظه‌ای کاهش یافت. نتیجه گیری: یافتههای این پژوهش نشان داد، ویروس نوترکیبAd5TATE(wt) قادر است به ‌صورت اختصاصی سلولهای سرطانی رده سلولی آدنوکارسینوما معده (AGS) را آلوده و پس از تکثیر، آن ها را متلاشی کند. لذا میتوان از ویروس Ad5TATE(wt) به‌عنوان عامل انکولیتیک علیه سلولهای سرطانی رده سلولی آدنوکارسینوما معده(AGS) استفاده کرد.                                                                                                                                                                      Aim and Background: Oncolytic viral therapy is known as promising approach for cancer treatment. Oncolytic viruses like adenoviruses can proliferate in cancerous cell and disintegrated these cells.  Serotype 5 adenovirus attached on surface of host cell via coxsackie-adenovirus receptors. The expression rate of this receptor is highly variable in surface of cancerous cells. The aim of the present study was production of an oncolytic adenoviruses with high selectivity feature for gastric adenocarcinoma cells (AGS) that can proliferate in this cell and destroy them. Material and methods: In this experimental study, the octreotate sequence was cloned into HI-loop of the adenovirus fiber. Thus, the pAd5TATE (wt) and pAd5TATE (ΔE1, ΔE3) recombinant vectors were constructed. pAdZ-TATE (wt) vector was synthesized using pADZ cloning system. Ad5TATE (wt) and Ad5TATE (ΔE1, ΔE3) recombinant viral particles were synthesized and propagated in HEK293 cells. Using the calcium phosphate procedure, the HEK293 cell line was transfected with the recombinant vectors and the new virus particles were produced. The AGS cell line was transfected by the Ad5 (wt) and Ad5TATE (wt) and the cell destruction of these cells were investigated in different times. Using the MTT test the cell surveillance rate was evaluated in AGS cell line that treated with recombinant vector. Cell destruction effects (CPE) started in less than 18 hours and were completed 72 hours after infection. Results: The titer of recombinant viruses was calculated by the TCID50 method as 107 (for 100 Lµ of viral solution). Also, Ad5TATE (ΔE1, ΔE3) virus titer was 107 TCID50/ml by the TCID50 method. The results showed that Ad5TATE (wt) could infect AGS cells compared to Ad5(wt). The results of the MTT test showed that with the increase in the concentration of viral particles and storage time, the number of live AGS cells decreased significantly. Conclusion: The findings of this research showed that the Ad5TATE (wt) recombinant virus could specifically infect gastric adenocarcinoma (AGS) cancer cells and destroy them after replication.
