fa بررسی پیام رسانی در سیستم ایمنی ذاتی مرتبط با siRNA از دیدگاه مولکولی Investigation of Innate Immune Signaling Concerned with siRNA from a Molecular Viewpoint ایمونولوژی Immonology مقاله مروری Review Article RNA interference) RNAi) که ازسوی siRNA) Small interfering RNA) میانجیگری می شود، ابزار قدرتمندی برای مهار بیان ژن در رده های مختلف سلولی است. siRNA ها علاوه بر عمل خاموش کردن ژن در القای سایتوکاین های پیش التهابی (Pro-inflammatory) و اینترفرون نوع I نیز دخالت دارند. گیرنده های ایمنی شناسایی کننده RNA شامل ( Toll-like receptor (TLR ها و پروتئین های سیتوزولی شناساگر RNA مانند: PKR1, RIG-I 2, MDA-5 3, 2'- 5'OAS4, IRF5, NF-κB6 می باشند. القای بیان اینترفرون ناشی از مسیرهای پیام رسانی است که به TLRها مربوط می شود. هرکدام از این رسپتورها در پاسخ به لیگاند های مختلف، توانایی القای پیام را دارند و سبب آغاز پاسخ سایتوکاین های پیش التهابی می شوند. در این مقاله درباره خاموش شدن ژن توسط siRNA، نقش فاکتورهای سلولی در تنظیم بیان ژن های سیستم ایمنی ذاتی و القای اینترفرون توسط siRNA دو رشته ای و تک رشته ای توضیح داده می شود. RNA interference (RNAi) has emerged as a powerful tool to suppress gene expression in a variety of cell lines by mediated small interfering RNAs (siRNA). In addition to the gene-silencing activity of siRNA, it can induce pro-inflammatory cytokines and type I interferon. RNA-sensing immonoreceptors include Toll-like receptor (TLR) family and cytosolic RNA-sensing proteins like Protein Kinase R (PKR), 2 َ -5 َ Oligo adenylate synthetase (2›-5›OAS), Retinoic-acid Inducible Gene –I (RIG-I), Melanoma Differentiation-associated gene-5 (MDA-5) and Nuclear Factor kappa B cell (NF-κB). Induction of IFN can be raised the signaling pathways associated with the TLR. Each of receptors group is able to induce signals in response to a variety of ligands, initiating the pro inflammatory cytokines response.
