سنتز و مقایسه دو آنالوگ ایرانی پپتید آزاد کننده گاسترین جهت تصویربرداری از تومورهای سرطانی سینه و پروستات

<b>
سابقه و هدف:</b> بومبزین پپتیدی است که تمایل بالایی به گیرنده های پپتید آزاد کننده گاسترین (بومبزین) دارد. تومورهایی همچون تومورهای پروستات، شش، پستان و کولون تعداد زیادی گیرنده پپتید آزاد کننده گاسترین را بیان می کنند. در این مطالعه، تولید و ارزیابی دو آنالوگ جدید نشاندار شده پپتید آزاد کننده گاسترین با تکنسیوم-99m و گالیوم-67 انجام پذیرفته است.
<div style="direction: rtl"><b>مواد و روش ها: </b>ترکیبهای DOTA-GABA-Bombesin (7-14) NH2، HYNIC-GABA-Bombesin (7-14) NH2 با استفاده از استراتژی Fmoc استاندارد تهیه گردید. تحلیل رادیوشیمیایی ترکیبهای نشاندار شده توسط روش HPLC انجام شد. پایداری پپتیدهای نشاندار در حضور سرم انسانی (دمای 37 درجه سانتیگراد) انجام و بررسی درون روی هر دو ترکیب نشاندار بر روی رده سلولی PC-3 انجام پذیرفت. </div><div style="direction: rtl"><b>یافته ها:</b> نتایج پایداری نشان داد که هر دو ترکیب دارای پایداری خوبی در سرم انسانی بوده و نتایج درون روی نشان داد که ترکیب 67Ga-DOTA-GABA-Bombesin (7-14) NH2 دارای درون روی بهتری نسبت به 99mTc-HYNIC-GABA-Bombesin (7-14) NH2 می باشد. بررسی توزیع بیولوژیکی نیز نشان داد که ترکیب 67Ga-DOTA-GABA-Bombesin (7-14) NH2 دارای نسبت تومور به کلیه و تومور به پانکراس بهتری نسبت به ترکیب دیگر است. همچنین سرعت دفع 67Ga-DOTA-GABA-Bombesin (7-14) NH2 از خون بهتر از ترکیب دیگر است. </div><div style="direction: rtl"><b>نتیجه گیری:</b> با مقایسه شرایط دو ترکیب می توان بیان نمود که ترکیب 67Ga-DOTA-GABA-Bombesin (7-14) NH2 دارای شرایط بهتری نسبت به 99mTc-HYNIC-GABA-Bombesin (7-14) NH2 می باشد.

Aim and Background:</b> Bombesin is a peptide showing high affinity for the gastrin releasing peptide receptor (GRPr). The prostate, lung, breast and colon tumors express many GRP receptors. The aim of this research was to compare two Iranian radiolabeled BBN analogs based upon the DOTA and Hynic chelators which could be used as a tool for the diagnosis of GRP receptor-positive tumors.
<div><b>Material and Methods:</b> Two peptides were synthesized using a standard Fmoc strategy which lead to produce DOTA-GABA-Bombesin (7-14) NH2 and HYNIC-GABA-Bombesin (7-14) NH2. Peptide conjugate showed good stability in the presence of human serum (37 degree centigrade). The internalization rates were studied in GRP receptor expressing PC-3 cells. Biodistribution of radiopeptide was studied in nude mice bearing PC-3 tumor.
</div><div><b>Results:</b> The results of internalization sowed that [67Ga]-DOTA-GABA-Bombesin (7-14) has a more internalization relative to 99mTc-HYNIC-GABA-Bombesin (7-14) NH2 in three stages. The results of biodistriution showed that the Tumor/Kidney and Tumor/Pancreas ratio after for [67Ga]-DOTA-GABA-Bombesin (7-14) was better than the other one. These data show that two bombesin analogs are specific radioligand for gastrin-releasing peptide receptor positive tumors and is a suitable candidate for clinical studies.
<div><b>Conclusion: </b>The results of blood clearance and internalization showed that the [67Ga]-DOTA-GABA-Bombesin (7-14) has good conditions as an imaging agent relative to the other one.
