fa شناسایی داروهای مکمل گانودرما لوسیدوم به روش PCR با استفاده از مارکرهای نواحی فاصله ساز داخلی ریبوزومی Identification of Ganoderma lucidum reishi in herbal supplements by PCR using markers of ribosomal internal transcribed spacer (ITS) region میکروبیولوژی Microbiology مقاله مروری Review Article سابقه و هدف: قارچ گانودرما لوسیدوم(Ganoderma lucidum) از مهمترین قارجهای درمانی شناخته شده در ایران و جهان می‌باشد. در این مطالعه با توجه به عدم وجود دستورالعمل ثابت و مشخص برای کنترل کیفی مکمل های قارچی در کشورمان، روشی بر پایه بررسی مولکولی برای استاندارد سازی و کنترل کیفی آن ارائه گردیده است. مواد و روش ها: هشت نمونه از دو برند تجاری مکمل‌های قارچ لوسیدیوم از داروخانه ها تهیه گردید. آزمون PCR با استفاده از   DNA استخراج شده از کپسول ها انجام پذیرفت. سپس از محصول PCR توالی‌یابی صورت گرفت و نتایج حاصل از توالی یابی با Genbank  و پایگاه ژنی مورد مقایسه قرار گرفته و درخت فیلوژنی مبتنی بر ITS آنها با تمامی توالی های شناخته شده از ایران رسم گردید. یافته ها: استخراج DNA از کپسول ها برای تست PCR بهینه گردیدند و محصولات PCR در ژل آگاروز بر اساس اندازه های مورد انتظار مشاهده شد. Blast و رسم درخت فیلوژنی توالی ها مشخص کرد قارچ موجود در یکی از نمونه های تجاری با برچسب آن تطابق کامل داشته و گانودرما لوسیدیوم بود و برند دیگر این تطابق را نشان نداد و به جای گانودرما لوسیدیوم  از گانودرما رسیناسیوم (Ganoderma resinaceum)  استفاده شده بود. نتیجه گیری: با توجه به تفاوت های کمی و کیفی مقادیر ترکیبات موثر در نمونه های مکمل های قارچی مختلف موجود در بازار هنگامی که مواد تشکیل دهنده را نمی توان از نظر مورفولوژیکی یا بیوشیمیایی تأیید کرد، بهترین روش موفق و مقرون به صرفه استفاده از روشهای مولکولی ژنتیکی مبتنی بر DNA و تجزیه و تحلیل فیلوژنتیک برای احراز هویت گونه‌های قارچی هستند.   Aim and Background: Ganoderma lucidum is one of the most important therapeutic mushroom known in Iran and the world. In this study, due to the lack of standard and specific instructions for quality control of mushroom supplements, a molecular method based on Internal Transcribed Spacer (ITS) for detection of mushroom species in supplements has been presented. Material and methods: Eight samples of two commercial brands of Ganoderma lucidium mushroom supplements were prepared from pharmacies. PCR test was performed using DNA extracted from capsules. Then, sequencing was performed from PCR product and the results of sequencing were compared with Genbank and their phylogenetic tree was plotted with all known sequences from Iran in Genebank. Results: DNA extraction from the capsules for PCR were optimized and PCR products were observed on agarose gel coordinated to the expected sizes. Blast analysis and phylogenetic tree of the sequences showed that the mushroom in one of the commercial samples was in full accordance with its label which was Ganoderma lucidum and the other did not show this conformity and was Ganoderma resinaceum instead of Ganodema lucidum. Conclusion: Due to the large differences in the quantity and quality of effective compounds in the various mushroom supplements available in the market, if the ingredients cannot be morphologically or biochemically confirmed, the best successful and cast effective method would be molecular method based on mushroom DNA and phylogenetic analysis to specify it species.   قارچ گانودرما لوسیدوم, واکنش زنجیرهای پلیمراز, نواحی فاصله ساز داخلی ریبوزومی, درخت فیلوژنی,Iau Science.
