fa بررسی تأثیر ایمونوتوکسین نوترکیب حاوی سم ریسین بر روی بیان ژن‌های آپوپتوزی BAX و BCL-2 در رده‌های سلولی سرطان کلورکتال و پستان The effect of a recombinant immunotoxin containing ricin toxin on the expression of apoptotic genes BAX and BCL-2 in colorectal and breast cancer cell lines فیزیولوژی Physiology مقاله پژوهشی Research Article سابقه و هدف: بیماری سرطان به طور عمده بهدلیل اختلال در تنظیم مرگ سلولی برنامهریزیشده است. از طرفی بیان بیش از حد<a href="#_ftn1" name="_ftnref1" title="">[1]</a> EGFR بر روی سطح بسیاری از سلولهای بدخیم ثابت شده است. از اینرو ایمونوتوکسین<a href="#_ftn2" name="_ftnref2" title="">[2]</a> بر علیه EGFR، که حاوی یک آنتیبادی یا یک قطعه از آنتیبادی متصل شده به یک مولکول سمی است، از قابل اعتمادترین راهبردها برای درمان سرطان است. در اینجا ما تأثیر یک مولکول جدید ایمونوتوکسین حاوی سم ریسین<a href="#_ftn3" name="_ftnref3" title="">[3]</a> بر روی سطح mRNA دو مولکول اصلی آپوپتوز در ردههای سلولهای سرطانی را بررسی کردیم. مواد و روش ها<a>:</a>  تأثیر مولکول href="#_msocom_3" id="_anchor_3" language="JavaScript" name="_msoanchor_3" on<illegal tag>mouseout="msoCommentHide('_com_3')" on<illegal tag>mouseover="msoCommentShow('_anchor_3','_com_3')">[M3]</a>   ژنهای پروآپوپتوتیک <a>BAX</a><a class="msocomanchor" href="#_msocom_4" id="_anchor_4" language="JavaScript" name="_msoanchor_4" on<illegal tag>mouseout="msoCommentHide('_com_4')" on<illegal tag>mouseover="msoCommentShow('_anchor_4','_com_4')">[M4]</a>  و آنتیآپوپتوتیک BCL-2 و ژن گیرنده EGF، در رده سلولهای سرطان پستان MDA-MB-468 و کلورکتال HCT116 با استفاده از <a href="#_ftn4" name="_ftnref4" title="">[4]</a>(qRT-PCR) انجام شد. از گلیسرآلدئید 3- فسفات دهیدروژناز بهعنوان ژن مرجع استفاده شد. یافته ها:  <a>تیمار</a><a class="msocomanchor" href="#_msocom_5" id="_anchor_5" language="JavaScript" name="_msoanchor_5" on<illegal tag>mouseout="msoCommentHide('_com_5')" on<illegal tag>mouseover="msoCommentShow('_anchor_5','_com_5')">[M5]</a>  با ایمونوتوکسین موجب افزایش معنیدار (05/0>P) بیان ژن BAX  و کاهش بیان ژنهای EGFR و BCL-2  و القاء آپوپتوز شد. همچنین سم ریسین باعث کاهش معنیدار BCL-2 و عدم افزایش معنیدار BAX در سلولهای سرطانی و کاهش معنیدار EGFR تنها در سلولهای سرطان پستان شد. بخش scFv<a href="#_ftn5" name="_ftnref5" title="">[5]</a> تغییرهای معنیداری بر بیان ژنها نداشت. نتیجه گیری: نتایج نشان داد که پروتئین نوترکیب پانیتومومب-ریسین، میتواند کاندید مناسب ایمونوتوکسین؟!!</div> </div> </div> Aim and background: Cancer is mainly caused by defect in natural regulation of programmed cell death (PCD). On the other hand, it is proved that, the EGFR is overexpressed on the surface of many malignant cell. Therefore, immunotoxin against EGFR, which consisting an antibody or fragment of an antibody linked to the toxic moiety is one of the most reliable strategies for cancer treatment. Here we evaluate the effect of a novel immunotoxin molecule harboring ricin, as a toxic moiety on the mRNA level of two main apoptotic molecules on cancerous cell lines. Materials and method: The Effects of immunotoxin molecule and it’s components including ricin toxin and antibody fragment on the mRNA level of pro- apoptotic BAX and anti-apoptotic BCL-2 and EGF receptor genes on the breast MDA-MB-468 and colorectal HCT-116 cancer cell lines were assayed by qRT-PCR. We used glyceraldehyde 3-phosphate dehydrogenase (GAPDH) as a reference gene. Results: Treatment with immunotoxin significantly increased the expression of BAX gene and decreased the expression of EGFR and BCL-2 genes (P<0.05) and induced apoptosis. Also, ricin toxin caused a significant decrease in BCL-2 and no significant increase in BAX in cancer cells and decreased EGFR only in breast cancer cells. The scFv part had no significant changes on genes expression.
