fa معرفی miR-200c به عنوان کاهنده بیان PD-L1 در رده های سلولی سرطان پستان Introduction of miR-200c as an attenuator of PD-L1 expression in breast cancer cell lines فیزیولوژی Physiology مقاله پژوهشی Research Article سابقه و هدف: گیرنده 1 پروتئین مرگ برنامهریزی شده سلول(PD-L1) ، به عنوان یکی از مهم ترین مولکول های کنترل کننده سیستم ایمنی در انواع سرطان ها (از جمله سرطان پستان)، نقش بسزایی در سرکوب سیستم ایمنی علیه سلول های توموری ایفا می کند. miRNA ها، به عنوان ابزارها و اهداف درمانی امیدوارکننده، نقش مهمی در کنترل بیان PD-L1 دارند. . در این مطالعه، با توجه به نقش احتمالی سرکوبکنندگی تومور miR-200c در سرطان پستان، مشارکت این miRNA در کاهش بیان PD-L1 و در نهایت کاهش تکثیر سلولی در سلول های سرطانی پستان بررسی شده است. مواد و روشها در این مطالعه آزمایشگاهی، ابتدا هدفگیری miR-200c بر روی3’UTR ژن PD-L1 با استفاده از نرم افزارهای بیوانفورماتیک و سپس با روش سنجش دوگانه لوسیفراز تایید شد. میزان بیان miR-200c و PD-L1 در ردههای سرطانی پستان نسبت به رده غیرسرطانی پستان اندازهگیری شد. پس از ترانسفکشن miR-200c به درون ردههای سلولی سرطان پستان، جهت بررسی اثر miR-200c بر میزان بیان PD-L1، روش qRT-PCR به صورت کمی مورد استفاده قرار گرفت. سپس برای بررسی میزان تکثیر سلولی، آزمون MTT انجام گرفت. یافتهها: در مطالعه حاضر، با استفاده از نرم افزارهای بیوانفورماتیک و سنجش لوسیفراز دوگانه، منطقه 3’UTR ژن PD‐L1 به عنوان یک هدف برای miR-200c شناسایی و تایید شد. متعاقباً، نتایج ما نشان داد که کاهش بیان miR-200c در سلولهای ردهی سرطان پستان با افزایش بیان PD-L1 ارتباط مستقیم دارد -0.68 , P value₌ 0.0158) =(r. همچنین افزایش بیان miR-200c در سلولهای رده سرطان پستان MDA-MB231، BT549 و MCF7، بیان PD-L1 را به میزان (به ترتیب 217/0±3/2، 021/0±11/2 و 012/0±146/0) کاهش داده است. بیان افزایش یافته miR-200c میزان تکثیر را در MDA-MB231 به میزان 012/0 ± 45/0 ، BT549 به میزان 2/0 ± 256/0 و در MCF7 به میزان 02/0 ± 164/0، در مقایسه با سلولهای ترانسفکت نشده کاهش داد. (میانگین ± SE) نتیجه گیری: یافتههای ما نشان میدهد که miR-200c با هدفگیری مسیر PD-1/PD-L1 و کاهش میزان بیان PD-L1 و نیز کاهش تکثیر سلول های رده سرطانی پستان، ممکن است بتواند به عنوان یک عامل درمانی برای جلوگیری از پیشرفت سرطان پستان باشد.   Aim and Background : PD-L1, one of the most important immune-regulating molecules in all cancers including breast cancer, suppresses the immune system against tumor cells. MiRNAs control PD-L1 expression as therapeutic tools and targets. This study investigated the role of miR-200c in reducing PD-L1 expression and breast cancer cell proliferation. Materials and Methods: In this study, miR-200c targeting the PD-L1 3'UTR was confirmed by bioinformatics and dual-luciferase assay. PD-L1 expression in breast cancer tissues and cell lines was checked compared to normal tissues and cell lines. qRT-PCR was used to measure PD-L1 expression after miR-200c transfection into breast cancer cell line. The MTT test was used to determine the effect of miR-200c introduction on cell proliferation.  Results: The 3'UTR region of the PD-L1 gene was identified as a target for miR-200c using bioinformatics software and a dual luciferase assay. The decrease in miR-200c expression in breast cancer cells is directly related to the increase in PD-L1 expression (r =-0.68, P value₌ 0.0158). Also, increasing the expression of miR-200c in MDA-MB231, BT549, and MCF7 breast cancer cells decreased the expression of PD-L1 by (0.217±2.3, 0.021±2.11, and 0.012±0.146, respectively). The increased expression of miR-200c decreased the proliferation rate in MDA-MB231 by 0.45 ± 0.012, BT549 by 0.256 ± 0.2, and in MCF7 by 0.164 ± 0.02, compared to non-transfected cells. (Mean ± SE) conclusion: MiR-200c may be able to prevent breast cancer progression by targeting the PD-1/PD-L1 pathway, reducing PD-L1 expression, and reducing breast cancer cell proliferation.
